Acetaminophen (APAP), also known as paracetamol, is a mainstream antipyretic and analgesic raw material worldwide and is listed in the pharmacopoeias of various countries. The synthesis of acetaminophen from p-toluidine is a classic, established industrial route comprising four main stages: acetylation for amino-group protection, oxidation and decarboxylation of the methyl group, hydrolysis to produce p-aminophenol (PAP), and final acetylation and purification to yield acetaminophen. Its advantages are evident: the technology is mature, and the barrier to entry regarding equipment investment is low. However, the process imposes strict quality requirements on the upstream p-toluidine, allowing for minimal impurity levels. The presence of heavy metal impurities in p-toluidine causes PAP to oxidize and darken, directly resulting in the final acetaminophen product failing to meet color specifications (making the quality of upstream raw materials a critical control point in this production route).
