We are Yixing Wencheng Chemical Co., Ltd., which has been focusing on the research, development, production and sales of methyl aniline series and derivatives for 40 years. As one of the earliest manufacturers of p-nitrotoluene in China to operate on a large scale, we have long served customers in the fields of pesticides, dyes, pharmaceuticals, and polyurethane materials, accumulating a wealth of first-hand application experience. Today, focusing on the p-nitrotoluene-to-folic acid industrial chain, we would like to share the practical and valuable insights we have gathered over the years. These experiences cannot be of great value if they remain internally, and we hope they can provide inspiration and assistance to our colleagues in the industry.
Folic acid, also known as vitamin B9, is a water-soluble B vitamin. As a key coenzyme in the human body's one-carbon metabolism, it participates in DNA and RNA synthesis as well as amino acid methyl conversion. Clinically, it is mainly used to prevent and treat megaloblastic anemia caused by folic acid deficiency; folic acid supplementation during preconception and early pregnancy can reduce the risk of neural tube defects in the fetus and can also help intervene in H-type hypertension, also known as hyperhomocysteinemia. In addition, folic acid is widely used in food fortification, dietary supplements, and multivitamin preparations.
p-Nitrotoluene is an upstream starting material in the folic acid synthesis route. It does not directly participate in the cyclization reaction of folic acid, but is mainly used to construct the para-aminobenzoyl structural unit required for the folic acid molecule. The reaction sequence is as follows: the methyl group of p-nitrotoluene is first oxidized to a carboxyl group, yielding p-nitrobenzoic acid, which is then converted into p-nitrobenzoyl chloride via treatment with a chlorinating agent. This acid chloride intermediate undergoes amide condensation with L-glutamic acid, followed by the reduction of the nitro group to produce N-(p-aminobenzoyl)-L-glutamic acid. This key intermediate subsequently undergoes a cyclization reaction with 2,4,5-triamino-6-hydroxypyrimidine to ultimately form folic acid, with the p-substituted benzene ring backbone being retained within the folic acid molecular structure.
As is well known, pharmaceutical-grade synthesis routes have much higher quality requirements for p-nitrotoluene compared to general industrial-grade routes. The folic acid pharmaceutical synthesis route employs stringent quality control standards for p-nitrotoluene. The main content must be ≥99.5%, and isomer impurities such as o-nitrotoluene and m-nitrotoluene must be strictly controlled. Once these isomers are introduced into subsequent processes, they will generate positional isomer impurities that are difficult to separate and remove, directly causing the folic acid product to exceed the relevant substances standard, and at the same time increasing the risk of genotoxic impurities. Moisture content must be ≤0.1%, and there are strict constraints on low-boiling and high-boiling impurities, as well as dinitro-compounds. Heavy metal ions, such as iron and copper, must also be strictly controlled; these ions can catalyze oxidation and accelerate side reactions, while also posing a risk of heavy metal residues in the final product. Furthermore, peroxide content must be monitored to ensure color stability. This raw material must also comply with GMP supplier audit requirements and full-lifecycle traceability protocols for hazardous chemicals. It should be emphasized here that o-nitrotoluene cannot be used for folic acid synthesis, as its ortho-substituted structure cannot construct the para-benzene ring unit required for the folic acid molecule, and it is strictly prohibited from being mixed into the folic acid raw material system. The p-nitrotoluene produced by Yixing Wencheng Chemical Co., Ltd. fully meets the requirements for dye and pharmaceutical grades in terms of purity, moisture content, and impurities.
In the actual large-scale production of folic acid synthesis, special attention needs to be paid to temperature, light, acid and alkaline environment, etc. Folic acid is highly sensitive to light, high temperatures, and extreme pH conditions; therefore, the entire industrial production process must be conducted under light-shielded (dark) conditions. The cyclization reaction requires precise control over pH and temperature—typically maintained in a weakly alkaline environment with the temperature strictly regulated between 35°C and 55°C. If the temperature exceeds this range, the folic acid core is prone to ring-opening, leading to the formation of pteridine-based degradation impurities. The feed ratio of 2,4,5-triamino-6-hydroxypyrimidine to N-(p-aminobenzoyl)-L-glutamic acid must be precisely managed. During the acylation and reduction stages, particular attention should be paid to controlling residual solvent levels. In the acylation and reduction stages, solvent residue levels must be carefully controlled. Crude products are generally refined using an acid-solution-alkali-solution recrystallization process, combined with activated carbon decolorization to effectively remove colored impurities. The reaction mother liquor cannot be recycled indefinitely, as impurities will accumulate; consequently, it must be periodically removed, separated, and recovered or treated. Nitro-containing intermediates are toxic; therefore, the relevant production stages must be operated within a closed system under negative pressure, with robust leak-prevention measures in place. Given the high salt content in the production wastewater, systems such as MVR (mechanical vapor recompression) evaporation and resin-based decolorization/recovery processes are required to facilitate resource recovery and ensure environmental compliance. During the final drying stage, temperatures must be kept moderate to prevent the degradation of folic acid and the darkening of its color. The impurity profile is continuously tracked throughout the entire production process, with a focus on monitoring genotoxic and degradation impurities.
Folic acid is a mature active pharmaceutical ingredient with stable and slow-growing demand. Its demand is driven by maternal and child health care, medication for chronic diseases such as H-type hypertension, mandatory global fortification of staple foods, and significant increases in dietary supplements and high-end formulations of active folic acid (5-methyltetrahydrofolate calcium). For manufacturers, the primary concerns are the long-term, stable supply of upstream raw materials—specifically p-nitrotoluene and p-nitrobenzoic acid—as well as the ability to strictly control impurities and ensure that the product consistently meets the standards set by the European Pharmacopoeia (EP) and the United States Pharmacopeia (USP).
